Modern bases of the theory of tertiary tooth bud development and its significance in pediatric dentistry

Congenital or acquired tooth loss remains a significant problem in modern dentistry, particularly in childhood, where it negatively affects the development of the maxillofacial system. In recent years, research in regenerative dentistry has focused on exploring the potential for biological tooth restoration. The aim of this study is to analyze the role of the TRG-035 drug and the USAG-1 protein in tooth regeneration, as well as to evaluate their significance in the formation of third dentition. The objectives include studying the biological function of the USAG-1 protein, analyzing the mechanism of action of TRG-035, and summarizing existing experimental findings. Material and method: This study was conducted as a systematic literature review. Scientific publications from PubMed, Scopus, and Web of Science databases published between 2000 and 2025 were analyzed. Relevant studies were selected based on predefined criteria and subjected to qualitative analysis. The results demonstrated that inhibition of the USAG-1 protein leads to activation of BMP and Wnt signaling pathways, resulting in the development of latent tooth germs. In animal models, the application of TRG-035 resulted in the formation of new functional teeth. In conclusion, the TRG-035-based approach represents a promising direction in tooth regeneration and may expand the possibilities of biological treatment of adentia in the future.
